Yes. First, you'll need to apply the diffuse map as polypaint data on the model. Then go to the Mask menu and under Mask by Color choose Mask by Intensity.
However, if you have a Black and White image, you could just convert it to greyscale and load it as an alpha map. Then in the Mask menu choose Mask by Alpha.
